Hands-on Training Programme on Bioprocessing
The workshop aims to offer a broad overview on various relevant techniques required for biopharmaceutical process development and optimization. Bioprocess development and optimization proceeds using an integrated approach of upstream processing, harvest technologies, downstream processing and analytical characterisation. Currently, there is an increased demand for scientists trained in bioprocess technology who can translate new discoveries into industrial-scale production and work at the interface of biology and engineering science.
This two-day hands-on training workshop shall be organized by the DBT Centre for Biopharmaceutical Technology at IIT Delhi. The workshop shall provide hands-on training in various relevant techniques required for biopharmaceutical product development in the following areas:
Module 1: Upstream Processing for Biopharmaceuticals
Module 2: Harvest Technologies for Biopharmaceuticals
Module 3: Downstream Processing for Biopharmaceuticals
Module 4: Analytical Characterization for Biopharmaceuticals
Applicants should select the desired module in which they wish to receive training while filling the application form.
The two breakthroughs of 20th century, recombinant DNA technology and hybridoma technology, have served as the genesis of the modern biotech era. Large-scale production of protein and protein based drugs are based on these technologies. Recombinant protein production has provided an approach to manufacture therapeutic and prophylactic protein drugs, which have been used to save uncountable lives from deadly and incurable diseases. Among microbial hosts, E. coli is the commonly used industrial host system for heterologous protein production. This two-day course work organized by the DBT Center for Biopharmaceutical Technology and National Biopharma Mission at IIT Delhi will broadly cover important microbial upstream topics including cloning, expression and production of bio therapeutics in E. coli and will also include hands on training on some of the key techniques.
As we move into the century of biology, the two breakthroughs hybridoma technology and recombinant DNA technology have served as the genesis of the modern biotech era. Harvest technologies serve as a bridge between upstream and downstream units, thereby enabling efficient large-scale production of recombinant protein. Type of clarification technique would depend on the type of expression system used (E. coli/CHO/P. pastoris). Techniques like Acoustic Wave Separator (AWS) and centrifugation help to remove cell debris from the protein rich supernatant and in cases of intracellular expression systems, these enable separation of IBs obtained post cell lysis. This two-day course work organized by the DBT Center for Biopharmaceutical Technology and National Biopharma Mission at IIT Delhi will broadly cover important cell clarification topics including E. coli cell lysis, IB extraction, protein estimation, clarification using Acoustic Wave Separator technology and will also include hands on training on some of the key techniques.
Bio-therapeutics forms an important part of modern medicines. Complexity of bio-therapeutics molecules necessitates elaborate purification processes involving a number of unit operations like chromatography, refolding, precipitation, filtration to target process and product related impurities. Selection of such unit operations depends on the expression system and specific purification requirements of the molecule. For example- refolding is required for proteins expressed as inclusion bodies whereas precipitation for removal of process related impurities like host cell proteins and DNA. Each unit operation demands a large number of parameters to be fine-tuned. This has led to the widespread adoption of high-throughput screening (HTS) technologies, which are characterized by miniaturization, automation and parallelization. This two-day course organized by DBT Center for Biopharmaceutical Technology and National Biopharma Mission at IIT Delhi will broadly cover important bio-pharma topics including high‐throughput techniques, chromatography, and filtration and refolding for purification of therapeutic proteins and will also include hands on training.
Development of protein based medicines is the current burgeoning segment of the global pharmaceutical industry. India has successfully placed itself as a leader in “generic” segment and is now steadily working its way up to make a global mark in the “biotherapeutic” segment as well. Unlike a “generic” drug that is a chemically synthesized identical copy of the innovator drug, production of biotherapeutics is a complex process as it involves recombinant expression of the specific protein/ peptide in a biological host. Therefore, the amount and type of permissible structural and functional variations have to be strictly controlled to avoid adverse immunogenic responses arising from the different proteoforms. This requires extensive physiochemical characterization of the structure, structural variation and impurities that may arise from the manufacturing process itself or the host used for the protein production. The potential for the presence of multiple variants in protein-based pharmaceuticals highlights the indispensability of state of the art analytical techniques that can measure these variants in a reliable and accurate manner. As no one technique can capture all possible structural variants of a protein in a high-throughput manner, multiple methods are used to study the different characteristics of each protein. This two-day course work organized by the DBT Center for Biopharmaceutical Technology and National Biopharma Mission at IIT Delhi broadly cover the various aspects of Analytical Characterization of Biotherapeutics including hands-on-training on the cutting edge techniques used in the industry as well as academia.
